3.1.4. Conclusion
Advantages of flow programming are compelling reasons to eliminate limitation of SI method, caused by mixing based on mutual penetration of sample and reagent zones. Because stacking sample with reagent zones in single channel forms a multicomponent gradient, that yields complex response, it is difficult to optimize assay protocol and it limits use of SI method for single and two reagent based assays.
There are two ways how to eliminate limitation imposed by single line SI manifold:
- By including a stirred chamber
- By including a confluence point
The first approach has proven to be efficient and found several applications (Section 3.1.3.). The second approach is the centerpiece of this Tutorial (Section 3.2.1.).
