Drug Resistance

Drug resistance belongs to the main obstacles emerging during the cancer chemotherapy. By the conversion of active drugs into corresponding metabolites, some biotransformation enzymes significantly contribute to this phenomenon. Using molecular cloning techniques, we generate vectors coding carbonyl reducing enzymes with potential role in the resistance of cancer cells to anthracycline treatment. These vectors are employed in cellular transfection and cytotoxicity experiments to demonstrate the participation of chosen carbonyl reducing enzymes on drug resistance. In addition, we investigate possible induction of examined carbonyl reducing enzymes following anthracycline treatment using qPCR and Western blotting methods. Such mechanisms can be targeted with specific inhibitors opening the way toward more efficient anticancer treatment.
