Purification and Characterization of Membrane Bound Carbonyl Reducing Enzymes from Human Tissues

We already developed and optimized solubilization of human liver microsomes, first purification step based on ion exchange chromatography and second purification step based on hydrophobic interactions chromatography. Now, we are focusing on affinity separation based on molecular recognition of carbonyl-reducing enzymes with xenobiotic oracin as a ligand. This compound is attached to the surface of magnetically active microparticles. Such carrier provides required parameters: proper selectivity and specificity enabling to isolate the target enzyme in sufficient quantity, purity, and activity.
